Airway mucin secretion is important pathophysiologically and as a model of polarized epithelial regulated exocytosis. We find the trafficking protein, SNAP23, selectively expressed in secretory cells compared with ciliated and basal cells of airway epithelium by immunohistochemistry and FACS, suggesting that SNAP23 functions in regulated but not constitutive epithelial secretion. Heterozygous SNAP23 deletant mutant mice show spontaneous accumulation of intracellular mucin, indicating a defect in baseline secretion. However mucins are released from perfused tracheas of mutant and wild-type mice at the (...) same rate, suggesting that increased intracellular stores balance reduced release efficiency to yield a fully compensated baseline steady state. In contrast, acute stimulated release of intracellular mucin from mutant mice is impaired whether measured by a static imaging assay 5 min after exposure to the secretagogue ATP or by kinetic analysis of mucins released from perfused tracheas during the first 10 min of ATP exposure. Together, these data indicate that increased intracellular stores cannot fully compensate for the defect in release efficiency during intense stimulation. The lungs of mutant mice develop normally and clear bacteria and instilled polystyrene beads comparable to WT mice, consistent with these functions depending on baseline secretion that is fully compensated. Social norms – rules governing which behaviours are deemed appropriate or inappropriate within a given community – are typically taken to be uniquely human. Recently, this position has been challenged by a number of philosophers, cognitive scientists, and ethologists, who have suggested that social norms may also be found in certain non-human animal communities. Such claims have elicited considerable scepticism from norm cognition researchers, who doubt that any non-human animals possess the psychological capacities necessary for normative cognition. However, there is (...) little agreement among these researchers about what these psychological prerequisites are. This makes empirical study of animal social norms difficult, since it is not clear what we are looking for and thus what should count as behavioural evidence for the presence (or absence) of social norms in animals. To break this impasse, we offer an approach that moves beyond contested psychological criteria for social norms. This approach is inspired by the animal culture research program, which has made a similar shift away from heavily psychological definitions of ‘culture’ and to become organized around a cluster of more empirically tractable concepts of culture. Here, we propose an analogous set of constructs built around the core notion of a normative regularity, which we define as a socially maintained pattern of behavioural conformity within a community. We suggest methods for studying potential normative regularities in wild and captive primates. We also discuss the broader scientific and philosophical implications of this research program with respect to questions of human uniqueness, animal welfare and conservation. (shrink)

Humans routinely modify their walking speed to adapt to functional goals and physical demands. However, damage to the central nervous system often results in abnormal modulation of walking speed and increased risk of falls. There is considerable interest in treatment modalities that can provide safe and salient training opportunities, feedback about walking performance, and that may augment less reliable sensory feedback within the CNS after injury or disease. Fully immersive virtual reality technologies show benefits in boosting training-related gains in walking (...) performance; however, they lack views of the real world that may limit functional carryover. Augmented reality and mixed reality head-mount displays provide partially immersive environments to extend the virtual reality benefits of interacting with virtual objects but within an unobstructed view of the real world. Despite this potential advantage, the feasibility of using MR-HMD visual feedback to promote goal-directed changes in overground walking speed remains unclear. Thus, we developed and evaluated a novel mixed reality application using the Microsoft HoloLens MR-HMD that provided real-time walking speed targets and augmented visual feedback during overground walking. We tested the application in a group of adults not living with disability and examined if they could use the targets and visual feedback to walk at 85%, 100%, and 115% of each individual’s self-selected speed. We examined whether individuals were able to meet each target gait speed and explored differences in accuracy across repeated trials and at the different speeds. Additionally, given the importance of task-specificity to therapeutic interventions, we examined if walking speed adjustment strategies were consistent with those observed during usual overground walking, and if walking with the MR-HMD resulted in increased variability in gait parameters. Overall, participants matched their overground walking speed to the target speed of the MR-HMD visual feedback conditions. The percent inaccuracy was approximately 5% across all speed matching conditions and remained consistent across walking trials after the first overall walking trial. Walking with the MR-HMD did not result in more variability in walking speed, however, we observed more variability in stride length and time when walking with feedback from the MR-HMD compared to walking without feedback. The findings offer support for mixed reality-based visual feedback as a method to provoke goal-specific changes in overground walking behavior. Further studies are necessary to determine the clinical safety and efficacy of this MR-HMD technology to provide extrinsic sensory feedback in combination with traditional treatments in rehabilitation. The Alternative Model of Personality Disorders integrates several theoretical models of personality functioning, including interpersonal theory. The interpersonal circumplex dimensions of warmth and dominance can be conceptualized as traits similar to those in AMPD Criterion B, but interpersonal theory also offers dynamic hypotheses about how these variables that change from moment to moment, which help to operationalize some of the processes alluded to in AMPD Criterion A. In the psychotherapy literature, dynamic interpersonal behaviors are thought to be critical for identifying (...) therapeutic alliance ruptures, yet few studies have examined moment-to-moment interpersonal behaviors that are associated with alliance ruptures at an idiographic level. The current study examined the concurrent and cross-lagged relationships between interpersonal behaviors and alliance ruptures within each session in the famous Gloria films. Interpersonal behaviors as well as alliance ruptures were calculated at half minute intervals for each dyad. We identified distinct interpersonal patterns associated with alliance ruptures for each session: Gloria ’s warmth was positively related with withdrawal ruptures concurrently in the session with Carl Rogers; Gloria’s dominance and coldness were related with increased confrontation ruptures in the session with Fritz Perls concurrently, while her coldness was also predicted by confrontation ruptures at previous moments; lastly, both Gloria’s dominance and Albert Ellis’s submissiveness were positively related with withdrawal ruptures. These interpersonal patterns demonstrated the promise of using AMPD dimensions to conceptualize momentary interpersonal processes related to therapy ruptures, as well as the clinical importance of attuning to repetitive, dyad-specific interpersonal cues of ruptures within each session. We identify participant-centricity and trustworthiness as the most important features of an MIC and discuss the implications for those seeking to create a sustainable, useful, and widely available collection of linked resources for research and other purposes.

One reason for its elusiveness is the “remarkable elasticity” (7) it has shown over time. Theological liberalism “supports critical engagement with both Christian traditions and contemporary intellectual resources” (6). In a way different from traditional theology, it “has been characterized by an affirmation of personal and collective experience, systemic social analysis and open theological inquiry” (6).At its best, liberal theology, appealing to Jesus’s ministry and especially his teaching of the reign of God, inspired people to believe they could make a difference in the here and now. Stereology is a valuable tool for neuroscientists, allowing them to obtain 3-Dimensional information from 2-Dimensional measurements made on appropriately sampled sections. This 3-D information is invaluable in correlating structural/functional relationships in the pursuit of far greater understanding of the function of the central nervous system. However, in carrying out such measurements, often based on limited data sets, there is a risk of experimenter bias. An important feature of modern design based stereology is to be aware of potential sources of bias (...) and eliminate them during the data collection. With many of the major neuroscience journals now insisting that quantitative data be presented, there is a greater need than ever for neuroscientists to understand the theory and practice behind quantitative methods, such as those offered by stereology. Phenomenological psychology is shown as a means to examine implications of mass-commodity culture, through the presentation of a phenomenological analysis of a TV commercial. This advertisement plays upon the vicissitudes of fathers' experiences of their relationships with their pre-pubescent daughters. The findings disclose an image of a father's ambivalently lived inability to tolerate his daughter's first sexual attraction to another male, and his attempt to continue to control the satisfaction of his daughter's bodily desire through commodities. This paper focuses on the “anti-realist” position of Newman and other artists for whom the Holocaust necessitated a renewed aniconic tendency in Jewish aesthetics. This chapter outlines the utility for employing case study methodologies to provide sufficient external validity upon which to craft policy relevant to maintaining healthy democratic politics. The broader theoretical context is an investigation into the conditions that might structurally condition democracies to fail via democratic means. Venezuela’s democratic decline serves as the basis for the heuristic case study, wherein the objective is to identify the failures of the Venezuelan case in a larger framework that addresses the complexity of institutional design (...) in democratic political systems states broadly. Cases are selected based upon the objective of the researcher, and similarly, the case study methodology chosen rests firmly on their research goals. Lastly, the chapter outlines the research design necessary for satisfying broader inquiry within the more modest approach to how heuristic case studies can be used to inform both theory and policy. It may be true that we are epistemically in the dark about various things. Does this fact ground the truth of fallibilism? No. Still, even the most zealous skeptic will probably grant that it is not clear that one can be incognizant of their own occurrent phenomenal conscious mental goings-on. Even so, this does not entail infallibilism. Philosophers who argue that occurrent conscious experiences play an important epistemic role in the justification of introspective knowledge assume that there are occurrent beliefs. (...) But this assumption is false. This paper argues that there are no occurrent beliefs. One of the greatest challenges facing materialist theories of the human mind is the problem of intentionality. As many non-materialists of various stripes have pointed out, it is very difficult to say, if the human mind is a purely material thing, how this material thing can be about or represent another thing wholly distinct from itself. However, for their part, these same non-materialists have relied heavily or exclusively on this intuition that one material thing cannot be about another. In this (...) paper, that hole in the intentionality objection to materialist theories of mind is filled by way of providing the germ of a new theory of the metaphysics of intentionality. On this view, intentionality just is partial identity. Partial identity is a concept introduced by David Armstrong and used by Donald Baxter to give a metaphysics of instantiation. The aboutness exhibited by our mental activity is simply a partial identity relation between the mind and the object of thought. This thesis, I contend, is a faithful interpretation of Brentano’s “intentional inexistence,” which the objects of thought have in the mind and which he held to be characteristic exclusively of mental phenomena. Some interesting and fortuitous consequences, including an elucidation of precisely why one material thing can never be about something distinct from itself, are then examined, and finally a defense against some objections is given. Embryo adoption, when oriented to the rescue of a dignified human person, is a merciful and morally licit response to an evil consequence of in vitro fertilization and the freezing of embryos. Those who object to embryo adoption not only misconstrue the relevant moral reasoning but exhibit confusion among the object, intention, and circumstances and between two very different potential objects. Because the mercy and charity effected through embryo adoption are at the very heart of moral action, juridical arguments that (...) undermine people’s confidence in these priorities have far-reaching, harmful implications. The objective list theory of well-being holds that a plurality of basic objective goods directly benefit people. These can include goods such as loving relationships, meaningful knowledge, autonomy, achievement, and pleasure. The objective list theory is pluralistic (it does not identify an underlying feature shared by these goods) and objective (the basic goods benefit people independently of their reactive attitudes toward them). In this paper, I discuss the structure of this theory and show how it is supported by people's considered (...) judgments. I then respond to three objections. First, I argue that there is no conceptual reason to favor a monistic theory of well-being over a pluralistic one (such as the objective list theory). Second, I argue that states of affairs can benefit people even though they hold no positive reactive attitudes toward them. And, third, I argue that objective list theorists can identify a fairly-determinate list of basic goods. (shrink)

This paper examines neuroethics as a discipline in which ongoing formation and development in both ethics and medicine are shedding new light on the care of patients diagnosed as being in a vegetative state. From the perspective of the Catholic moral tradition, the author proposes that ethics and recent developments in functional neuroimaging form a complementary relationship that gives rise to an ethical imperative: because we can care for patients in a vegetative state, we should do so. This imperative for (...) care finds a particular expression in the ethical debate in light of the 2004 statement of Pope John Paul II that assisted nutrition and hydration for patients in a vegetative state should be considered, in principle, ordinary care. What do neurology and developments in scientific research teach us about patients in a vegetative state? During the 12th century, certain questions came into focus and inspired speculation about the heavens and the earth, namely, “mobile being”. In other words, nature was “discovered” in the 12th century. It is in the wake of this discovery that, in the 1250s, Bonaventure developed his view of the created world while commenting on the interpretation of the Genesis account of creation found in the church fathers, as these had been anthologized in Peter the Lombard's Sentences.

Bonaventure holds that God does all things with power, wisdom, and goodness; such as in the case of the restoration. If Incarnation is examined as a work of God in the light of power, wisdom, and goodness, we can see why it is the most perfect of all God's works, for there cannot be any greater act of power than to combine within a single person two natures: the human and divine. While Bonaventure stresses the gratuity of the redemption — (...) that is, God was in no way obliged to save us — he is very clear that man could not have been saved by any healer other than God. Bonaventure uses “sacrament” to refer to all signs of faith in the Redeemer, even those that are not explicitly focused on Jesus of Nazareth. He refers to this as the “diversity” of the sacraments. “Sacraments” in this sense were instituted from the very beginning, but they have enjoyed diversity through three different ages and their concomitant laws: the law of nature, the law of scripture, and the law of grace.

Ectogestation involves the gestation of a fetus in an ex utero environment. The possibility of this technology raises a significant question for the abortion debate: Does a woman’s right to end her pregnancy entail that she has a right to the death of the fetus when ectogestation is possible? Some have argued that it does not Mathison & Davis. Others claim that, while a woman alone does not possess an individual right to the death of the fetus, the genetic parents (...) have a collective right to its death Räsänen. In this paper, I argue that the possibility of ectogestation will radically transform the problem of abortion. The argument that I defend purports to show that, even if it is not a person, there is no right to the death of a fetus that could be safely removed from a human womb and gestated in an artificial womb, because there are competent people who are willing to care for and raise the fetus as it grows into a person. Thus, given the possibility of ectogestation, the moral status of the fetus plays no substantial role in determining whether there is a right to its death. (shrink)
